CSols, Inc. Enhances and Extends GenoLogics Clarity LIMS for Adaptive Biotechnologies

NEWARK, Delaware – CSols, Inc. the premier Laboratory Informatics consultancy, announced the successful completion of the enhancement and extension of GenoLogics Clarity Lab Information Management System (LIMS) for Adaptive Biotechnologies, the leader in high-throughput immunosequencing. To make use of several new features, and to further automate the processing and management of its assays, Adaptive Biotechnologies decided to upgrade its GenoLogics Clarity LIMS. In scoping the requirements for this upgrade, Adaptive Biotechnologies realized that some enhancements, extensions, and interfaces (instruments and systems) were needed, and turned to CSols, a Clarity Certified Application Development (CCAD) partner, for assistance.

Adaptive engaged CSols to develop the enhancements, extensions, and interfaces, because of CSols’ expertise with Clarity LIMS, the Clarity LIMS API, and its laboratory and domain knowledge.

The adoption of Electronic Laboratory Notebooks (ELNs) in both academic and commercial labs continues to grow in the market.  In fact, the ELN market growth rate has been clearly outpacing that of a Laboratory Information Management System (LIMS) in many market segments.  Informatics systems vendors have noticed this for some time and have moved to take advantage of this buying trend by developing and bringing to market new ELN offerings at a record pace.

A quick, informal count of the ELN vendors shows greater than 50 commercial vendors with an additional half dozen or so Open Source solutions being offered.  Of these, many are designed to be generic systems that can be configured to satisfy the needs of many lab environments while others are purpose built to provide a narrow and deep solution for a particular environment.  And let’s not forget about Laboratory Execution System (LES) solutions which are designed to automate and implement bench level control in labs.  With such a broad choice in ELNs the question of “Which is the best ELN?” is continually being raised.  We would love to answer this question for you once and for all but unfortunately the answer is “It Depends!”

5 Tips for Choosing the Right Informatics Solution

Not the answer you were necessarily looking for, but the only truthful answer despite what various ELN companies would tell you.  What the Best ELN for you depends on a multitude of factors, or what most in the industry would term, your business needs and system requirements.  Some examples of these include:

  • What type of lab are you running? QA/QC?  R&D?
  • What type of science are you performing? Small Molecule?  Biologics?
  • Do you need to interface to other systems? ERP? MES?
  • Will you be integrating to instruments or instrument systems?
  • Are you in a regulated environment or not? Pharma?  Academia?
  • Do you have an IT infrastructure to support the ELN?
  • Do you have staff to support the ELN?

When you answer all these questions and many, many more then you can determine what the Best ELN is for your organization.

I know that this appears very intimidating, if not downright impossible, but do not despair.  There are informatics consulting organizations that can help.  When selecting a consulting organization to assist you in selecting the best ELN for your laboratory, you should ensure that they are:

  • Experts in ELN and ELN selection (get references)
  • Domain experts (i.e. your industry and your type of lab)
  • IT experts
  • Regulatory experts (if your lab is in a regulated environment)
  • ELN vendor neutral

Utilizing SOPs to fill Computer System Validation (CSV) Gaps

SOP-Gap-BlogHave you ever been working on the Computer System Validation (CSV) of a Laboratory System and hit a brick wall?  The validation comes to a standstill because the software/hardware does not meet one of three main aspects of computer system validation testing:

  1. Does not function as intended
  2. Does not meet an aspect of 21 CFR Part 11
  3. Does not meet a general CSV requirement

After struggling with the system on your own and contacting the vendor, it is soon determined that an alternate solution must be found in order to meet the regulations.  The quality organization of your company will be a key driver in determining what will be an appropriate solution.  And if sufficient in-house CSV expertise does not exist, it is highly recommended that a laboratory software validation Subject Matter Expert (SME) be engaged.

So how can you overcome this challenge?  The primary way is to hopefully find an “engineering solution” wherein the software can be configured/customized to solve the challenging issue.  When this is not possible a procedural solution must be found that is acceptable to the business, the IT organization, and the quality approver for the validation.  Let’s take a closer look at the three challenges described above.

Does Not Function as Intended

The most difficult issue to overcome is when the software/hardware does not function as intended.  This is the rarest of the issues since one would hope that the software vendor has tested their system for intended use and remedied any issue found.  However, bugs and function failures still do occur.

The first way of addressing this issue is to contact the vendor to determine if there is a known solution.  If there is not, your next course of action is to attempt to turn off that functionality in the software.  If both of these approaches fail then you must first show due diligence by performing the following actions:

  1. Document in the CSV documentation that the function will not be used
  2. Document and train the users that the function will not be used
  3. Create an SOP that documents that the function will not be used for GxP purposes.

During data review for this system once it is validated, the document reviewer also needs training and a procedure to verify that this function was not used by an analyst.

Does Not Meet a 21 CFR Part 11 Requirement

In the current compliance environment, data integrity has become very important.  Companies must not only show that they are in control of all data that is created, but also must show that the raw data is inalterable and fully audit trailed.  They must show that there is accountability for that data from its creation to its ultimate archiving.  The metadata for these system must also be under control.  If electronic signatures are used by the company, they must show that the signatures are attributable to an individual.

Laboratory instrumentation that have a computer and store data must meet these criteria.  In many cases the vendors have created these instruments and the associated software with the intent to meet 21 CFR Part 11, but in many cases there are gaps or non-compliance issues.  These situations can make it difficult for a laboratory to successfully complete their CSV project.

The key to overcoming these challenges is to set the laboratory system up as a hybrid system.  Certain aspects of the 21 CFR part 11 portion of the validation can be met by the software but others can then be met by procedure (i.e. SOPs).  The way that a company combines the paper procedural SOPs with components of the electronic system yields the compliance that is required.

Remember, when gaps are first identified, a risk assessment should be performed to identify and determine the overall risk and exposure.  This will help guide how and what extent of testing and documentation will be required to satisfy compliance.

Does Not Meet a Computer System Validation Requirement

A computer system validation requirement such as: backup, windows security, recovery, etc. are common requirements that have issues passing CSV.  This could be for a variety of reasons.  Some software applications may stop running if they become securely locked down in the operating system.  Others will not be able to backup all files since some are “open files”.  In these instances, a procedural solution combined with a software solution may be required.  This will require the attention of the business, the vendor and the IT department to solve.

Solving these issues can be complex.  It is critical to have people that are experienced in the system, compliance and IT solutions in order to address these issues.  All these disciplines bring a view that will be able to create a solution that can meet the scrutiny of the agency looking at compliance and data integrity.

11/9/15 Pharmaceutical Company contracts CSols for Empower 3 Validation

A specialty pharmaceutical company is in process of upgrading their Waters Empower 1 CDS to Empower 3. For this project, the client required a Software Validation Consultant to review the current validation lifecycle documentation for Empower 1 and provide guidance for the upgrade. CSols was chosen for their Domain and Regulatory expertise, as well as their deep experience working with Waters Empower. CSols will be supporting the validation activities by creating a Validation Assessment Report to identify the existing gaps in the Empower 1 validation and identify corrective actions required to validate Empower 3, based on industry best practices and GAMP 5 guidelines for a configurable system.

Kyle McDuffie runs his Christiana-based consulting business, CSols Inc., with a high degree of trust.

His management philosophy? Provide employees with the resources they need to succeed, and trust them to get the job done right. And don’t make them work on Saturdays.

“When you’re supposed to be working, do your best work,” says McDuffie, president of CSols, explaining that he encourages workers to dedicate their time off to family or community pursuits. “When you’re not supposed to be working, don’t work. Spend time with your family. Volunteer. This whole work-life balance thing, for us, is actually real.”

It’s easy to see why McDuffie’s employees voted him a top leader of a Delaware-based small business, according to results of a Top Workplaces survey conducted for The News Journal by WorkplaceDynamics.

CSols is an $8 million business that helps Fortune 100 pharmaceutical manufacturers, chemical firms, forensic testing organizations and other clients implement complex information systems like LIMS, ELN, and other scientific software in their laboratories.

The firm has about 20 employees in Delaware – 47 total – that help by providing expert advice.

Many of CSols’ employees have worked in the industries for which they now consult, providing the company with a level of expertise that the company’s clients really appreciate, McDuffie says. McDuffie himself is a former Dow Chemical Co. chemist.

The LIMS, ELN and other laboratory software systems that CSols employees help implement track active ingredients in drugs, quality control in vaccine batches, evidence tracking in forensic labs and, in some cases, patient information.

McDuffie and Sandy Ahmed, vice president at CSols, have run the business since a 2004 management buyout from a U.K.-based parent.

Ahmed calls McDuffie a “servant leader,” and a “collaborator” who leads the company, while placing trust in the company’s employees.

McDuffie explains: “Life is a team game,” he said. “It’s hard to make your way through, whether it’s business or regular life, all by yourself.”

“Fundamentally you have to trust that people are going to do what they are supposed to do. You have to give them the tools. Then you have to come back and see that’s happening. But people usually enjoy that and respect that, that you’ve given them the trust that ‘hey, you know your job.’ ”
